Green, Reticulated, Spotted

This page serves as a repository of research notes on Toxin OE04, the fourth mutagen unlocked in Old Egypt by Renard. It uses 6 Green, Reticulated, Spotted (Negative to Crackly, Hairy, Phosphorescent) moss and 4 Hairy Tooth, 2 Heart of Ash, 3 Heaven's Torrent, 1 Spiderling, 7 Sun Star mushrooms.

First mutation test: L-Vampire R-Silken

On the Vampire, an IO has been replaced by an IY, making the outer stamen orange.

The range on the right splint is somewhat left of center based on where the Y has been removed from the Silken.

Second mutation test: L-Silken R-Vampire

An IO gene is now missing from Vampire. No change is visible on the Silken.

Right-Vampire 5 or 6

Next Step

Not much to go on yet, though the right splint has been narrowed down. While still facing so many unknowns, a useful double cross is Delicate-Fracture. Both have few genes with noticeable effect, and if we can determine exactly what gene is affected on the Fracture it will tell us what was affected on Silken too since they both have the same number of genes.

Third mutation test: L-Delicate R-Fracture

On the left, Delicate's dwarf allele has been disrupted so the mutagen is hitting far to the right on the left splint.

Fracture is now a Semidwarf, so a Giant allele has been broken and the mutagen affects the left part of the genome when the flower is on the right splint.

Fourth mutation test: L-Fracture R-Delicate

On the left, no change is apparent to the Fracture. That suggests the gene affected is in the GOR set of genes.

On the right, a Magenta outer stamen (IO) has been replace by Cyan (IR)

Discussion and Next Step

We've got a good feel for the left now based on the fourth test.

Left-Fracture 14(R)
Left-Silken 14(U)

Based on the range, that narrows down Left-Vampire to 9 or 10. From the results of the first test though:

Left-Vampire 10(O)
Left-Delicate 13(G)

What can we say about the right splint? The second test suggests that the gene affected is left of center, but not by far. That is rather restrictive on the fourth test, so:

Right-Delicate 6(O)

That limits Vampire to genes 4 or 5 and Silken/Fracture to 6 or 7. From the results of the first test, the range of Vampire was restricted to 5 or 6 so:

Right-Vampire 5(I)
Right-Silken 7(Y)
Right-Fracture 7(Y)

Excellent, now to move onto a larger genome. The ranges for all flowers up through Energy have been narrowed down to a two genes each, so let's compare that and a few more possible flowers.

Energy (Genes 14-33): GYYY GYYY GRRO GRRO UROR
Left-Splint genes: 32-33 Right-Splint genes: 15-16

Clarity (Genes 11-28): IY GORR GROR GRRO GORR
Left-Splint genes: 28
Right-Splint genes: 13-14
Left-Clarity, Right-Energy is not particularly informative.
Left-Energy, Clarity-Right would make it clear whether it's the 13th or 14th gene affected on Clarity, but would not give any information on Energy if it's the 13th gene.

Blush (Genes 6-22): GOOO V UROO V IO V IOO V
Left-Splint genes: 20-21
Right-Splint genes: 9-10
Left-Energy, Right-Blush would not be informative on which Energy gene is affected if it is the 10th gene of Blush affected.
Left-Blush, Right-Energy would let me know which gene was affected on the left splint, but not the right. All possibilities have a unique result when crossed with Clarity however.

Crown (Genes 13-29): V UR V OO V UR V OR V UROR
Left-Splint genes: 28-29
Right-Splint genes: 13-14
Not informative at all.

Fifth mutagen test: L-Blush, R-Clarity

Color missing from entire stamen on Blush, Clarity has has a GORR allele broken.
Left-Blush 20(O)
Right-Clarity 13(G)

Sixth mutagen test: L-Clarity, R-Blush

Color broken on the petals of both Clarity and Blush.
Left-Clarity 28(R)
Right-Blush 10(V)

Discussion and next step

The range has now been narrowed enough that Morning can be characterized.

The range for Dusk and Crimson have been narrowed.

Putting a Y onto the left splint of crimson will work just fine for characterizing the effects, since we'll break the GROR allele and either get a Dwarf, nothing, or a Giant.
Crimson-Left, Energy-Right

Putting an R onto the right splint of crimson will also characterize the effects well.
Fracture-Left, Crimson-Right

Seventh mutagen test: L-Crimson, R-Energy

Crimson has remained Single-Dwarf. Left-Crimson 81Y

Eighth mutagen test: L-Clarity, R-Crimson

Nothing has changed on either flower. This is an unexpected result. Either I've got the ranges wrong, or the Crimson genome is listed wrong. If a UROO on the official genome is in fact a UROR, that would explain the trouble. I'll conduct more testing before concluding that the genome listing might be off.
